Muscarinic agonists
What are the direct agonists?
Carbachol
Metacholine
Butanechol: supraventricular tachycardia
Muscarine
Pilocarpine: Glaucoma
What are the indications?
Glaucoma
Supraventricular tachycardia
GI, urinary bladder atony: paralytic ileus, atonic bladder
Myasthenia gravis
Receptors
M1: Gq coupled receptors, activating PLC pathway, glandular secretion, CNS
M2: Gi coupled receptors, decreasing cAMP, decreasing intracellular Ca - In nodal cells of heart - negative heart effects
M3: Gq coupled receptors, PLC pathway, GI motility, endocrine and exocrine secretion, myosis, accomodation, urination - as smooth muscle there are going to be contracted , bronchooconstriction, bronchal gland secretions
What are the indirect agonists?
Cholineseterase inhibitors
Neostigmine
Physostigmine
Pyridostigmine
edrophonium
