Understanding Opioids
Mechanism of Action
Binding to these receptors lead to pain relief, euphoria and CNS depression
Opioid agonists inhibit neurotransmitter release & increase dopamine levels in the brain's reward system
Primarily act on 3 types of receptors; mu, delta & kappa, located in the brain and peripheral nervous system
Physiological effects vary based on type of opioid & specific receptor activated, influencing both therapeutic & harmful outcomes
Effects on the Body

CNS - causes sedation and euphoria but also respiratory depression, which can lead to overdose
Gastrointestinal Tract - slows down guit motility, leading to constipation, a common side effect
Chronic use leads to tolerance, where higher doses are needed to achieve the same effects due to receptor downregulation & desensitisation
Withdrawal symptoms include nausea, vomitting, diarrhea & psych distress indicating physical dependence
