Methods used to color microorganisms or cells
Make cells visible under microscope
Simple Staining
Uses only one dye (e.g., methylene blue, crystal violet)
Stains entire cell with one color
Helps in observing:
Shape of cells
Size of cells
Arrangement of cells
Differential Staining
Uses more than one dye
Differentiates between types of bacteria
Helps in classification of microorganism
Help in identifying structure, shape, and type of microbes
Gram Staining
3. Gram Staining
Most important bacterial staining method
Divides bacteria into two groups:
Gram Positive
Stains purple
Thick peptidoglycan cell wall
Gram Negative
Stains pink/red
Thin cell wall with outer membrane
Acid-Fast Staining
Used for bacteria with waxy cell
wall
Detects Mycobacterium species (e.g., TB bacteria)
Resistant to acid alcohol decolorization
