Staining & it's Types: Staining is a biochemical technique used to increase the contrast of microscopic images by applying dyes that bind to specific cellular components. Since most biological cells are transparent and lack color, stains allow scientists to visualize structures like nuclei, cell walls, and organelles.

This method involves the use of a single basic dye (like Methylene Blue, Crystal Violet, or Safranin) to color all cells the same way.
· Purpose: To observe the size, shape, and arrangement of cells.
Mechanism: Basic dyes have a positive charge that is attracted to the negatively charged surface of bacterial cells

These techniques use more than one dye to distinguish between different types of organisms or different parts of the same organism.

The most famous differential stain used to classify bacteria into two large groups based on cell wall composition.

Gram-Negative: Appear pink/red because they have a thin peptidoglycan layer and lose the primary stain, taking up the safranin counterstain instead.

Special Staining

These stains are designed to highlight specific parts of a cell that are not easily visible with standard methods.

· Capsule Staining: A "negative" staining technique where the background is stained, leaving the protective capsule as a clear halo around the cell.
· Flagella Staining: Uses a mordant to thicken thin hair-like flagella so they can be seen under a light microscope.
Negative Staining

Unlike other methods, the dye (such as India Ink or Nigrosin) is acidic and negatively charged.
· Mechanism: The cell repels the dye, leaving the organism colorless against a dark, stained background.
· Benefit: Since no heat-fixing is required, the cells remain undistorted, providing the most accurate representation of size.
