main examples of inflammatory disorder: IgE mediated allergic reactions, rheumatoid arthritis, sepsis
IgE mediated allergic reaction- immediate allergic reaction, common in atopy which a genetic tendency to allergy the main cell involved= mast cells. Main steps
-

-
-
-
Rheumatoid arthritis- autoimmune inflammatory disease, causes chronic joint inflammation, leads to cartilage and bone destructiion
sepsis- acute organ dysfunction caused by an abnormal response to infection, It is inflammation that has become dangerously dysregulated. Common infection sources pulmonary, abdominal, genitourinary, bloodstream, skin

-
what is inflammation? the body's response to injury, infection or tissue dammage
main triggers
-
-
-
-
-
Bacterial toxins, e.g. LPS.
-
-
Steroidal antinflammatory drugs examples: hydocortisone, prednisolone, dexamethasone
-
Inflammatory mediators: vasoactive mediators (histamine and kinins), lipid mediators (prostaglandins and leukotrienes), protein mediators (complement, cytokines e,g TNF, IL-1 IL-6, chemokines)
Histamine: released mainly by mast cells causes vasodilation, increased vascular permeability and helps produce redness, heat and swelling
prostaglandins: made from arachidonic acid and produced using COX-1 and COX-2 enzymes effects; vasodilation, enhance mediator action, cause fever, short lived
Leukotrienes: made from arachidonic acid produced by 5-lipoxygenase effect; attract neutrophils, increase leukocyte adhesion, activate macrophages
Kinins: produced when precursor proteins are acted on by kallikrein. Main examples; bradykinin, kallidin. Effects vasodilation, pain stimulation, increased vascular permeability, smooth muscle contraction
-
Possible outcomes of inflammation: full resolution- harmful stimulus removed and tissue returns to normal, partial resolution- some tissue damage remains can lead to chronic inflammation, Chronic inflammation- inflammation continues example; tuberculosis granuloma
