It's used with countable things in the plural. That is, things you can count one by one.

Affirmative

Example: They live in Madrid.
He / She / It + verb + -s / -es
Example: She lives in Madrid.
Negative
I / You / We / They + don't + verb (infinitive)
Example: They don't live in Madrid.
He / She / It + doesn't + verb (infinitive)
Example: He doesn't speak French
Interrogative
Do + I / you / we / they + Verb (infinitive) + ?
Example: Do you speak English?
Does + he / she / it + Verb (infinitive) + ?
Example: Does she speak English?
