-Gram stain: Gram negative

-the Combination of these test results confirm my unknown organism is Pseudomonas aeruginosa (Pa)

- I performed a nitrate test after the positive citrate test.
- The tube changed to red, that was an indicated that my organism was negative for nitrate reduction. My organism cannot reduce to nitrate to nitrite or any nitrogen gas.

Citrate test
- I use citrate as my test to determine if the bacterium can use citrate as its only carbon source.
-This test helps to separate bacteria look similar in another test.
- This test provides a clear negative or positive results.
Negative is (blue) and positive is (green)
- I think this test was perfect to narrow down unknown possibilities
The results came back positive! next Nitrate test
-After isolating colonies from the streak plate
-My Gram staining results helped me to find the cell wall structure and bacterial shape.
-The bacteria appeared Gram negative (pink)
-The shape was rod-shape
This result helped to separate the mixed culture from the broth that was given.
-Now that my unknown was isolated and characterized by gram stain, I could continue with biochemical testing. Next: Citrate test

- I used the Gram staining technique
-The microscopic examination showed purple (Gram-positive)
-Shape was cocci and the arrangement was long chains
-this help to guild me to the next step in which was the biochemical test.

- Gram staining revealed purple, gram positive. The shape I saw was Cocci in long chains.
-This was confirmation that my organism was Gram positive and now I can move on to biochemical testing. I choose MR test
