Definition and Concept

Therapeutic Index

- Therapeutic index is a comparison of effective versus lethal dose.
- Therapeutic index can be determined by LD50/ED50
TI50 HIGH, Safety HIGH
- A safer, more reliable, and thus TI in drug development:
TI = TD1 /ED99 or LD1 /ED99.
- Margin of Safety: LD1 / ED 99 or TD1 / ED 99
- Therapeutic Windows (Safety)

THERAPEUTIC WINDOW
- A dosage range of a drug to produce its therapeutic effect
• Between the minimum therapeutic dosage and the minimum toxic dosage
• E.g. min. therapeutic dose for drug A is 10mg/L, toxic dose is 25mg/L → Therapeutic window is 10-25mg/L
