cell fractionation
homogenisation- breaking up the cell, tissue cut up and kept in a buffered solution

ultracentrifugation- separating the organelles, homogenised tissue spun in an ultracentrifuge at a low speed for 10 mins, ultracentrifugal force causes a sediment containing the heaviest organelle to separate out leaving a supernatant (repeat until desired organelles separated out)
solution kept ice cold- not affect enzyme activity, no breaking down
isotonic solution- doesnt affect osmosis/ water moving in or out the sell/ prevent bursting or shrivelling

buffer is used- keep the pH of the solution the same to not affect enzyme activity- don't denature/ organelle structure remains same
