Administered orally except morphine, hydromorphone and oxymorphone (high first pass metabolism)

CNS (analgesia, euphoria, sedation, respiratory depression, antitussive, miosis, nausea + vomiting
Periphery (bradycardia (*merperidine causes tachycardia*), constipation, biliary contraction, urine retention (inc. sphincter tone)
MOA
Acts on inhibitor G protein receptors mu (analgesic, endorphins), kappa (analgesics, dynorphins) and delta (enkephalins bind) found in spinal cord and periphery.
