Wk1 Biochemistry & Basic Methods
Immune based assay methods
EMIT: Enzyme Multiplied Immunoassay Technique
PETIA: Particle Enhanced Turbidimetric Immunoassay
PETINIA: Particle Enhanced Turbidimetric Inhibition Immunoassay
ELISA: Enzyme-Linked Immunosorbent Assay
Methods used to measure various analytes
Potentiometry
Measurement of an electric potential between two electrodes
Absorbance Spectrophotometry
Measurement of light absorbance by a solution at one or more specific wavelengths, usually in the UV-VIS spectrum
Turbidimetry
Measures reduction in direct passage of light through a turbid (particulate containing) solution
Nephelometry
Measures amount of light scattered/reflected/refracted in a different direction to direct light path. Detector is placed at an angle to direct light path
Fluorescence Spectroscopy
Detection of a substance through its emission of light at one or more specified wavelengths after its absorbance of light a different (shorter) wavelength
Chemiluminescence
Light of a particular wavelength is produced by a substance due to the decay of an excited electron (that was excited by a chemical reaction) to a ground state
Electro-Chemiluminescence
A voltage trigger (removal of an e-) is used to produce a chemical reaction that excites a compound to emit light at a particular wavelength
Ion Selective Electrodes
Use a membrane with selective permeability to detect specific ions
Analytes
Ions, salts & minerals (e.g. Na, Ca, Cl, K, Mg, Fe)
Small organic molecules (e.g. glucose, bilirubin, cholesterol, aspirin, cocaine)
Large macromolecules (e.g. albumin, ALP, LD, Ig, lipoproteins)
Blood is either collected in a tube with:
An
anticoagulant
(e.g. EDTA or heparin), & thus fluid component is
plasma
Without an anticoagulant
then the fluid part is called
serum
