Cells Turned from purple (Strain A) to yellow (Strain B):
Plate should be disregarded as not reliable; possibly due to contamination
Cells stayed purple in both Strain A to Strain B:
Plate is reliable can carry on analysis
Look at wells containing 9-AA (positive control for TA154)
If a significant amount of the cells in the plate turned from purple in Strain A to yellow Strain B it indicates that a Frameshift mutation has occurred
Look at wells containing N4-AC (positive control for TA100)
If a significant amount of the cells in the plate turned from purple in Strain A to yellow Strain B it indicates that a Base pair mutation has occurred
Compound of interest caused both a frameshift and a base pair mutation
Look at wells containing the test compound
If most of the cells are yellow in strain B of the -S plate we can not determine whether the test compound requires activation to cause the mutations.
If most of the cells are yellow in strain B of the +S plate we can determine that the test compound requires activation to cause the mutations
If most of the cells are purple in strain B of the +S plate we can determine that the test compound does not require activation to cause the mutations
If a significant amount of the cells in the plate remained purple from Strain A to Strain B it indicates that no Base pair mutation has occurred
Compound of interest causes only a frameshift mutation
Look at wells containing the test compound
If most of the cells are yellow in strain B of the +S plate we can determine that the test compound requires activation to cause the mutations
If most of the cells are purple in strain B of the +S plate we can determine that the test compound does not require activation to cause the mutations
If most of the cells are yellow in strain B of the -S plate we can not determine whether the test compound requires activation to cause the mutations.
