Disease-modifying anti-rheumatic drugs
leflunomide
Mechanism of action: it archives its effects by inhibiting the mitochondrial enzyme dihydroorotate dehydrogenase (DHODH), which is essential in the de novo synthesis of uridine monosphate (rUMP) , which is needed for the synthesis of DNA and RNA
indications : it is used to treat rheumatoid arthritis which causes pain, swelling and loss of functions on the body's joints
contraindications: it is contraindicated in patients who are known with leflunomide hypersensitivity and teriflunomide hypersensitivity
Side effects: Dizziness, Hair loss, Headache, Heartburn, Sin rash, Hypertension, Low blood cell count
Storage Conditions: it should be stored in room temperature of 25 degrees Celsius
Route of administration: orally
Methotrexate
Mechanism of action: it works by slowing and stopping the expanding of cancer cells and suppresses the immune system.
indications: adults with severe, active, rheumatoid arthritis and children with polyarticular-course juvenile rheumatoid arthritis
Contraindications: should not be administered in daily doses. other contraindications are renal failure, liver disease, active infectious disease and excessive alcohol consumption
Side effects: abdominal pain, Hair Loss, Headache, Itching, Dry cough
Storage Conditions: must not be refrigerated, not exposed to heat and light, it should be stored at a cool dry and dark place.
Route: can be given by oral, subcutaneous, and intramuscular
Biological Response Modifiers
Abatacept
Mechanism of Action: it binds to the C80 and CD86 molecule, and prevents the second signal, the T cell can not be activated.
Indications: to treat adults with moderate to severe rheumatoid arthritis
Contraindications: Hepatitis, Hepatitis B, COPD, Tuberculosis
Side effects: Cough, Dizziness, Infusion reaction, Serious infections like Tuberculosis
Storage Conditions: the diluted solution must be stored at 2 to 8 degrees celcous
Route of administration: 30-minute intravenous infusion
Glucocorticosteroids
Prednisone
mechanism of action: it decreases inflammation via suppression of the migration suppression of the migration of polymorhonuclear leukocytes and reversing increased capillary permeability. it supresses the immune system by reducing the activity and the volume of the immune system
Indications: Endocrine Disorders, Rheumatoid Disorders, Opthalamic disorders, Respiratory Diseases.
Contraindications: active, untreated tuberculosis, herpes simplex infection, diabetes.
Side effects: Cataracts, Increased Cholesterol, Mood swings, High blood pressure
storage conditions: store away from light and heat. store at cool dark places
Route of Administration: taken orally, depending on the doctors prescription.
