Methylprednisolone
Contraindications
• it must not be used to patient with fungal infection,ulcerative colitis,liver disease,multiple sclerosis
• intramuscular corticosteroids are contraindicated for idiopathic thrombocytopenic purpura

Endocrine disorder for example congenital adrenal
hyperplasia,hypercalcemia associated with cancer and non suppurative thyroiditis.Allergic state for example bronchial
asthma,contact dermatitis and seasonal orperenni alallergic
rhinitis

Mechanism of action
methylprednisolone is a synthetic corticosteroid with anti-
inflammatory and immunomodulating properties.Unbound
glucocorticoids cross cell membrane and bind with high affinity to specific cytoplasmic receptors,modifying transcription and protein synthesis.By this mechanism,glucocorticoids inhibit leukocyte infiltration at the site of inflammation,interfere with mediators of inflammatory response,suppress humoral immune response.

Mechanism of action
prednisone is a glucocortocoid receptor agonist it is metabolized first in the liver it active form prednisolone. Prednisolone crosses cell membrane and bindbwith high affinity to specific cytoplasmic receptor.This inhibit leukocyte in filtration at the site of inflammation an suppress humonal immune responses and reduction of oedema or scar tissue.

Fludrocortisone
Contraindications
Fludrocortisone must not be given to patients with fungal
infection,thyroid disorder,renal impairment,active infection and those who are allergic to fludrocortisone acitate
Side effects
insomnia,eye bruising,increase hair growth,irregular menstrual periods or amenerrhoa,dizziness
Indication
prophylactic management of asthma,Addison'sdisease,
adrenogenetic syndrome,primary and secondary adrenocortical insuffieciency

Mechanism of action
The binding or activation of mineralocorticoids
receptors by fldrocortisone in turn causes an ion and water transport and thus raises extracellular fluid volume and blood
preasure and lowers potassim level.

Indication
Relief inflammatory and pruritic manifestations of corticosteroids-responsive(adrenal insufficiency).it is also used to treat immune and allergic disorder such as arthritis,lupus, severe psoriasis,severe asthma,ulcerative colitis and Crohn's disease

therapeutic Hydrocortisone is a synthetic or semi-synthetic
analog of natural hydrocortisone hormone produced by the
adrenal gland with primary glucocorticoid and minor
mineralocorticoids effect.As a glucocorticoid receptor agonist,hydrocortisone promotes protein catabolism,gluconeogenesis,capillary wal lstability renal excretion of calcium and suppress immune and inflammatory response.
