Temozolomide
Mechanism: is an alkylating agent used to treat glioblastoma. Alkylation (addition of methyl group) causes DNA damage (breakage of N-glycosidic bond). The body uses MGMT (methylguanine methyl transferase) to remove the methyl group from the N7 position of damaged guanine residues with allows the glycosidic bond to reform.
