EFFECTOR MECHANISM
CD8 CTL
- Activation of T cells: MHC-peptide recognized by T
- Tumor need to prod new epitope (neoepitope) by mutation/expression of novel
- For strong CD8/4 response: Neoepitope bind with high affinity to MHC (then induce 2nd signals)
- Ingestion of antigen by DAMP-activated DC lead to cross presentation of antigen on MHC 1
- Principal mechanism of adaptive immune protection against tumor is killing of tumor cells by CD8+ CTL
NK
- Kills cells that have reduced MHC 1 and express ligand for NK cell-activating receptor
CD4 T HELPER CELLS
- Providing cytokines for differentiation of naive CD8+ T into effector and memory CTL
- Secrete cytokine that increase expression of MHC 1
ANTIBODIES
- Ab kill tumor cells by activating complement or by ADCC, Fe receptor-bearing macrophages/NK mediate killing (passive immunotherapy)
CYTOKINES
- TNF-a and IFN-y have antitumor effect, upregulate MHC class 1 and 2 antigens on tumor
