Clinical Pharmacology II KPC10202
Sir Eddy Abdul Raub
Parasympathetic stimulation increases intestinal motility and GI secretions and relaxes sphincters
Antiemetics
-
Mechanism of Action
1.5HT3 Antagonist;it block the serotonin receptors in CNS and Gastrrointestinal tract so it can be used to treat post opp and chemotherapy drugs nausea or vomitting
2.Prokinetics;they block the dopamine neurotransmitter also they promote gastrointestinal motility and quick gastric emptying
3.Antihistamines;they block the histamine neurotransmitter and act by an effect on vomitting centre by producing sedation
4.Anticholinergics;it will block the neurotransmittter acetyl choline in central and peripheral nervous system

-
Laxative/Purgatives
-
Mechanism of Action
1.Osmotic laxatives draw water into the intestine to increase the mass of stool, stretching musculature which results in peristalsis
-
3.lubricant laxatives increase water retentionin the stool.prevent water absorption from the stool and lubricate as well as soften intestinal contents
4.Stool softener allow more fluid are fat to penetrate the faeces, producing a softer fecal mass
-
-
-
Antacids
-
Mechanism of action
They achieve their effects by neutralizing gastric acid, inhibiting gastric acid secretion or protect gastric mucosa
-
-
-
-
Proton Pump inhibitors
-
Mechanism of action
Inhibit proton pump which is final common step in gastric acid secretion. It also have antisecretory action.
-
-
-
-
Antidiarrheals
-
Mechanism of action
Antidiarrheals active opiods receptor in GI tract to decrease intestinal motility and to increase the absorption of fluid and sodium in the intestine
-
-
-
-
-
Use Histamine, other CNS depressants including opiods and sedative, hypnotic drugs causes additive CNS depression
-
-
Antacids decreased absorption of anticholinergics,sucralfate,h2 receptor antagonist etc..
-
-
Antacids may inhibit absorption of H2 receptors antaginists.Cigaette smokking increeases gastric acid secretion and may decreases the effectiveness of H2 receptors
