Immunoassays
Glossary of terms
Antibody = glycoprotein composed of heavy and light chains, recognises and binds antigen
-
-
-
Antibody-Antigen Binding
Binding occurs through weak non-covalent interactions (van der Waals, hydrogen bonding, ionic interactions )
Sum of interactions between epitope and Abs, short range and complementary of shape i.e. lock and key

Types of Immunoassays
-
-
-
Enzyme Immunoassay (EIA)
Reporter enzyme attached to either antibody or antigen most common format is ELISA (Enzyme-linked-immunosorbent assay)
Common ELISA formats - direct assay, indirect assay or capture assay 'sandwich'
Fluorescent Immunoassay
Uses a fluorescent label/product from enzymatic rxn and is more sensitive than colorimetric measurements used in EI (spectrophotometer)
Definition
-
Presence is denoted by an indicator reaction and its generally used to detect proteins, hormones or antibodies
