Methotrexate polyglutamates inhibit AICAR more than methotrexate in rheumatoid arthritis
inhibition leads to accumulation of AICART ribonucleotide
Nucleotide synthesis inhibition prevents cell division
Which then inhibit adenosine indeaminase
Enzymes inhibition is responsible for nucleotide synthesis
Causing adenosine triphosphate accumulation
It converted to methotrexate polyglutamate by polyglutamate when entering tissues
Stimulating adenosine receptors leading to anti-inflammatory action
