Chromatography
Analytical technique
According to IUPAC Chromatography is defined as: A method for the separation of the components of a sample ,in which the components are distributed between two phases, one of which is stationary while the other moves in definite direction(mobile phase)
From Greek: chromatos-color, graphein-to write
1903 Tswett-plants pigments seperated on chalk columns
1931 Lederer & Kuhn-LC of carotenoids
1938-TLC and ion exchange
1950 Reverse phase LC
1954 Martin & Synge(Noble Prize)
1965 instrumental LC (water)
Stationary phase: phase which remain fixed
Mobile phase: phase which move on stationary phase
Elution: Removal of absorbed species
Eluent: fluid entering column / solvent that carries the analyte
Eluate: mobile phase leaving the column
Chromatogram: The finished paper or thin layer plate having bands of color is called chromatogram.
Resolution: The degree of separation of compounds of similar characteristics.
