Resolution
the clarity of an image: the higher the resolution the clearer the image & the more detail you can see - the ability to distinguish between two objects hat are very close together
image = 2D, low resolution

vacuum
why: if there were any air molecules in the microscope the electrons would be disrupted by them & therefore not be able to create an image of the specimen
effects: can only use dead specimens cause living cells would explode in the vacuum (can view living in light microscope)

calibrating a microscope

eyepiece graticule - tiny scale inside eyepiece, divided up into eyepiece units. we need to find the value of each eyepiece unit to then find the size of the stuff were looking at.
stage micrometer - a scale of known length built into a slide, the entire scale is 1 mm (1000μm) & divided into 100 divisions, each off the smallest divisions is 10μm

e.g. if 1000μm on the stage micrometer = 40 eyepiece units (eu)
1eu = 1000/40 = 25eu
so each eyepiece unit is worth 25μm at the objective lens this was calculated at
