AUROCLAVE
PURPOSE
- to denature and coagulate the protein and induce lethal chemical changes within the micro-organism
- temperature of the autoclave chamber : 121 degree celcius
DISADVANTAGE
Property of HDPE: Tg value = -90 degree celcius, Tm value = 130 celcius Since the autoclave needs to be operated at 121 degree celcius, HDPE would become rubbery and melt. When it;s cooled it would wrap
ETO STERILIZATION
uses EtO Gas
alkylation of amine groups on nuclei acid which prevents normal cellular metabolism
DISADVANTAGE
-toxic, flammable and potentially carcinogenic
proper PPE
aeration of EtO to remove residual EtO in material after EtO sterilization
IONIZATION RADIATION
uses gamma ray / electron beam
- cause ionization of key cellular components -> death of micro-organism
DISADVANTAGE
chain scission of polymer
decrease the molecular weight -> decrease in mechanical property
discolouration of the polymer (not aesthetically pleasing)
SELECTION OF TECHNIQUE
EtO Sterilization
As autoclave would cause the HDPE packaging to warp making it not desirable.Among the 3 techniques, I would choose EtO Sterilization
.
I would not choose radiation sterilization as radiation sterilization would cause
chain scission
which would affect the mechanical properties of the packaging material. It would also cause the HDPE packaging to tern yellow, making it not aesthetically pleasent.
I would choose EtO sterilization as it uses EtO gas and water vapor to sterlize the material and also HDPE is permeable. Thus, it would be more desired as it can fully sterilize the material without affecting the mechanical property of the material. However, precaution would need to be taken as EtO gas are toxic and it must be clean properly before using it as it might cause reaction
