Place the slide on stage. Look through eyepiece Turn focus wheel to get a clear image Start with low objective lens. Increase magnification of lens and refocus
Take samples of your cell. Add small amount of chemical stain. Chemical stain help them to be more visible. Apply the cell on slide. Slower put the cover slip onto the slide. Avoid air bubbles
