Part 1: Sample Preparation
Mix the contents inside the serum sample by mechanical means (inversion or tapping
HDL Analysis: Pipette 200 μL of the serum into a microcentrifuge tube and label as HDL
Sample Processing: Add 10 μL of HDL reagent to the serum sample. Use a vortex mixer to mix
Incubate at room temperature for 15 minutes
Centrifuge for 15 minutes at 5000 rpm
Glucose Analysis: Pipette 3 μL of serum into a microcentrifuge tube and label as G
Insert analysis end (white) inside the device
Pipette 2 μL of serum onto a parafilm
Insert the sampling end of the test strip (yellow tip) into the serum
Read glucose concentration and record
Total Cholesterol Analysis: Pipette the rest into a microcentrifuge tube and label as T
T and HDL Measurement: Label 5 cuvettes with the following: B, LS, HS, T, and HDL
Add 2 mL of cholesterol reagent to each cuvette
B: Add only water for cuvette B
HS standard: Mix tube with vortex mixer and transfer 20 μL in to HS cuvette
Cover with parafilm and mix gently
Incubate for 15 to 20 minutes
Take absorbance reading of each cuvette using a spectrophotometer
Determine the concentration of Total cholesterol and HDL cholesterol using the given equations. Record data

LS standard: Mix tube with vortex mixer and transfer 20 μL into LS cuvette
HDL: Add 20 μL to HDL cuvette
T: Add 20 μL to T cuvette
