(Strain improvement techniques), Formation of new gene combinations among…
-
- Formation of new gene combinations among those present in different strain
- Recombinant used for both genetic analysis & strain improvement
- To generate new products
- Genetic Engineering technology
- Used to isolate & clone the genes of interest
- Isolated gene will be constructs using appropriate genetic tech in lab condition
- The isolated gene will be cloned into a vector (plasmid of a bacteria)
- The vector will expresses in bacteria
- Bacteria which successfully received the vector will expressed new gene - desired product
- This bacteria is known as recombinant bacteria
- rDNA tech is used to:
- Produce recombinant product
- in metabolic engineering
- Recombinant protein - The cloned new rDNA MO produced the desired protein. - Eg. insulin, interferon
- Metabolite engineering - The metabolic activities of an organism are modified by introduction of a transgene.
- affects the enzymatic, transport, regulatory functions of its cells
- Eg. high production of amino acid isoluccine in C. glutamicum & ethanol by E. coli
- Developing a suitable medium and refining or retuning the fermentation process can cause an increased yields of the product.
- The changes depends on the nature of the strain used for fermentation process
- Also depends on the strain maximum ability to synthesized the product
- Beyond particular level the strains productivity cannot be improved further - Major limitation of this approach
- Developing a suitable medium and retuning the fermentation can be done via:
- Modification of physical parameters - Temperature, Agitation
- Modification of chemical parameters - pH, Aeration
- Modification of biological parameters - Enzymes, Nutrients (C, N, minerals)
- Productivity of the organism is controlled by its genome and it is essential to modify the genome to increase the yield
- Genetic manipulation allows improving the productivity of the strain
- The culture requirements of the modified organism should be fully examined
- Mutagens can cause mutation in MO, it could cause a sudden heritable changes
