Penicillinase-resistant
Methicillin
- Highly penicillinase resistant, but NOT acid resistant
- Inducer of penicillinase production
- Adverse effects:
- Haematuria
- Albuminuria
- Reversible interstitial nephritis
- Largely replaced by Cloxacillin
Cloxacillin
- It has isoxazolyl side chain
- Highly penicillinase & Acid resistant
- More active than Methicillin against Staphyloccocci, but NOT MRSA
- incompletely but dependably absorbed for oral route
- Plasma protein bound >90%
- Elimination via kidney, liver
- Half-life: 1 hour
