Administration of the Antibody against Human CCR6 in the MMTV-PyMT Knock-in Mice for the Investigation of Mammary Tumorigenesis
Aims and Work program
Specific aims
- To develop a mouse model for studying the role of CCR6 in mice, anti-hCCR6 mouse Abs and hCCR6-Tg/mCCR6-/- MMTV-PyMT mice are generated and characterized. The model applies humanized mAB against hCCR6, making this model more relevant to human and provide values for pre-clinical study (include WB/Immunoprecipitation, chemokine assay to ensure that antibody connect to the receptor, validation compared with human)
- To examine whether anti-CCR6 influences in situ mammary tumorigenesis and TDLNs, Tg mice are intratumorally injected with or without anti-CCR6 [treating course?] at 8-week-old. Tumor growth curve, area of the hyperplastic lesion from HE staining, survival curve, cell proliferation (in vitro cell culture and Ki67 staining after intratumoral administration), macrophage attraction (flow cytometry, etc.) are going to be analyzed
- To investigate the combined therapy with taxanes to examine whether the drug can increase the expression of CCL20 and thus hinders the effect of the drug and whether ant-CCR6 can recover the drug's effect
