CHROMATOGRAPHY
Qualitative Analysis
- all about having a characteristic retention time of molecules
- when using same set of conditions, a given molecule will elute at the same time
- having peak at a retention time is not confirmation that the peak will represent the molecules, it can be other things as well that include to interfere the analysis
- the peak is also not a confirmation for the existence of the molecule
- the absence of peak indicates that the compound is either absent or below the detection limit (DL)
- chromatography not tells many things about molecules as it is just separation tool that need to be pair with other specific mode of detection
-
Quantitative Analysis
- having detector responding to eluting compound, use peak height / peak area to obtain amount of material
- peak area over peak height
:wavy_dash: as it not depends on broadening of the peaks
- perform a calibration curve with a series of different concentrations of standards
:wavy_dash: to get response of detector to pure compounds
- variation and sample volume inject
:wavy_dash: use internal standard
:wavy_dash: prevent imprcision in results as we use very small volumes in injection (20 microlitres)

Resolutions
:star: Common Resolution
:wavy_dash: the ideal resolution is between 1.5 and 10
:wavy_dash: resolution that is greater than 10 is wasting time and have no real benefits
:star: Ways to Improve
:wavy_dash: change the retention factor
:wavy_dash: changing the selectivity factor
:wavy_dash: increase N
