The epoxide side chain of mupirocin is similar to isoleucine and competes with it for binding sites on the synthetase enzyme

Bacterial protein and RNA synthesis are inhibited when mupirocin reversebly binds to bacterial isoleucyl-RNA synthetase

The cellular concentration of isoleucyl-tRNA becomes depleted
