Amylin analog pramlintide
Mechanism of action
affects glucose control through several mechanisms
slowed gastric emptying, regulation of postprandial glucagon, and reduction of food intake
Glucagon-like peptide-1 exhibits similar properties as amylin, with the exception of insulin secretory effects.
Storage
should be at room temperature before injecting
Refrigerate (2°C to 8°C; 36°F to 46°F)
Route of administration
subcutaneously immediately prior to each major meal
≥250 kcal or containing ≥30 grams of carbohydrate
dose should be administered subcutaneously into the abdomen or thigh
Indications
adjunctive treatment in patients with type 1 or type 2 diabetes
who use mealtime insulin therapy and who have failed to achieve desired glucose control despite optimal insulin therapy.
Contraindications
poor compliance with current insulin regimen.
poor compliance with prescribed self blood glucose monitoring.
have a HbA1c >9%
recurrent severe hypoglycemia requiring assistance during the past 6 months.
Side effect
nausea
vomiting
headache
hypoglycemia, if taken alongside insulin
