Nitrate Spectrophotometric Analysis - Method validation
pH
Required pH 8.5
Measure of solutions before and after the addition of buffer
LNSW vs Wilkins using default Cd method
Rationale: Cd reduces best at ph 8.5; Wilkins (DW) - slightly acidic
Try "Alkaline" water?
Use of LNSW - better buffering capacity of seawater
How to adjust the pH? Should we try to adjust it as well?
Which stage of the method will the pH be tested? Should we measure before and after addition of the buffer for all solutions?
Why not include the other matrices as well since we are not changing the Cd part of this methodology? We are essentially testing two different matrices. Unless we're doing extra steps to ensure pH is at 8.5?
Should we add a %recovery part?
Prepare extra reagents and working standards so we can test the pH at every step?
Matrix
LNSW
Distilled Water
Different brands?
Wilkins (WDW)
Milli Q (MQ)
Artificial Sea Water (ASW)
Cadmium amount
Reagents
Are we preparing the reagents in the same matrices? for each of the matrix tested? Or just in DW? or MilliQ?
Hindi na siguro noh? Kasi konti lang dinadagdag na reagents hehe (Lara)
Mixing time
Methodology
Marj and Ryan made 10 replicates - so dapat ba ung standards natin ay more than 100ml?
70 samples per matrix * 4 matrices = 280 samples total; prepare reagents for 350 samples (?)
Blank + 6 concentrations * 10 replicates = 70 samples
