Beta-Adrenergic Blockers
Nadolol
indication
the treatment of hypertension, either alone or in combination with other antihypertensive agents, especially thiazide diuretics
For the long-term management of angina pectoris.
mechanism of action
nadolol competes with adrenergic neurotransmitters (e.g., catecholamines) for binding at sympathetic receptor sites. Similar to propranolol and timolol, nadolol blocks sympathetic stimulation mediated by beta1-adrenergic receptors in the heart and vascular smooth muscle. Pharmacodynamic consequences of beta1-receptor blockade include a decrease in both resting and exercise heart rate and cardiac output, and a decrease in both systolic and diastolic blood pressure. Nadolol may reduce reflex orthostatic hypotension.
route of administration
Orally(Administer nadolol without regard to meals.)
storage conditions
Avoid excessive heat
Protect from freezing
Protect from moisture
Store at controlled room temperature
contraindications
AV block, bradycardia, cardiogenic shock, heart failure, hypotension, pheochromocytoma, pulmonary edema, sick sinus syndrome, vasospastic angina, ventricular dysfunction
Cerebrovascular disease
Diabetes mellitus
side effect
Dizziness, drowsiness, weakness, and cough may occur. If any of these effects persist or worsen,
propanolol
side effects
dry eyes
nausea and drowsiness
wheezing or symptoms of bronchitis
mechanism of action
Propranolol is a competitive antagonist of beta-1-adrenergic receptors in the heart. It competes with sympathomimetic neurotransmitters for binding to receptors, which inhibits sympathetic stimulation of the heart. Blockage of neurotransmitter binding to beta 1 receptors on cardiac myocytes inhibits activation of adenylate cyclase, which in turn inhibits cAMP synthesis leading to reduced PKA production.
contraindications
Reversible airways diseases, particularly asthma or chronic obstructive pulmonary disease (COPD)
Slow heart rate (bradycardia) (<60 beats/minute)
Atrioventricular block (second- or third-degree)
indications
Hypertension. Inderal is indicated in the management of hypertension. ...
Angina Pectoris Due to Coronary Atherosclerosis. ...
Myocardial Infarction. ...
route of administration
orally
intraveneously
storage
Store at room temperature away from light and moisture
