Mechanism of Action
Ketoconazole, fluconazole, and itraconazole block the activity of a sterol in a fungal wall.
Posaconazole, a new drug, inhibit the synthesis of ergosterol leading to inability of the fungus to form cell wall.

CONTRAINDICATIONS
Hepatic dysfunction. Ketoconazole, fluconazole, posaconazole, and terbinafine can cause serious hepatic toxicity.
Endocrine or fertility problems. Ketoconazole is absolutely contraindicated in patients with this condition because of its effects on these processes.
