Transcription
Elongation
transcription apparatus continues to unwind and denature the DNA in the 5’ direction while RNA polymerase adds nucleotides to the growing RNA in the 3’ direction
Termination
elongation ends when RNA polymerase transcribes the terminator sequence that causes the detachment of RNA polymerase from the DNA template
Rho-independent termination - Once transcribed, the inverted repeats form a hairpin loop that destabilizes RNA polymerase from the DNA
Hairpin Loop - unpaired loop of messenger RNA (mRNA) that is created when an mRNA strand folds and forms base pairs with another section of the same strand
Rho-dependent termination - Once the rut sequence is transcribed, Rho binds the terminator and uses energy from ATP to travel down the mRNA until it catches up with RNA polymerase. When Rho reaches RNA polymerase, it destabilizes RNA polymerase from the DNA
Initiation
transcription apparatus (RNA polymerase and accessory proteins) attach to the DNA at the promoter region and the double stranded DNA is denatured to form the transcription bubble
