ACICLOVIR- Against HSV-1/2 and VZV. Analogue of guanosine and there is an initial phosphorylation step by viral thymidine kinase which activates the drug.
MOA:It gets phosphorylated to triphosphate. It is incorporated into the DNA chain and so the chain is terminated. Aciclovir triphosophate is a direct inhibitor of viral DNA Pol.
Tx: HSV encephalitis, neonatal HSV, disseminated HSV, disseminated varicella zoster and severe varicella pneumonia, recurrent genital HSV infection, orolabial HSV infection, Herpes zoster and for prophylaxis of HSV infections in immunocompromised patients
Toxicity: Nephrotoxicity
