Lec 15: Clearing
-
Why?
-
raise refractive index of tissue, making it somewhat transparent
-
After clearing process, the tissues are more translucent and can be observed clearly using microscope
-
-
Procedure
Hard Tissues
1) •Hard tissues are to be transferred from 100% dehydrant to the 3:1 mixture of dehydrant and clearing agent (Xylol).
-
3) Finally, the material is changed in 100% clearing agent twice or thrice
Soft Tissues
-
In the final stage
if the clearing agent is not foggy, dehydration is complete
If the clearing agent is foggy, dehydration is not complete
material is to be transferred to 100% dehydrant and once complete the material be brought to clearing agent
A clearing agent has to have ability to remove dehydrating agent from the tissue and also to be dissolved in paraffin wax medium
