Neurons in substantia nigra gradually die and dopamine production is reduced. Dopamine is not enough, basal gangla doesn't work properly tp control body movements.

Causes of PD

Drug-induced
Antipsychotic drugs that block dopamine receptors, eg. haloperidol

Reactive free radicals (H2O2) from the catabolism of dopamine, causing cell death.

Difficult to initiate movement. Muscles become stiff, making movement jerky
