Mechanism of Action

The increase in cGMP that is produced by the release of nitric oxide result in a decrease in platelet aggregation
Nitroglycerin produces a dose related dilation of both arterial and venous bed
Relaxes smooth muscle of epicardium coronary arteries and reduces coronary artery spasm.
Decrease smooth muscle contractile result from lowering of intracellular calcium and desensitatization of myosin light chain (a contractile protein) to the effect of calcium
Contraindication
Type 5 PDE inhibitors potentiate the action of nitrates used for angina: 24hours of taking sildenafil is likely to produce hypotension.
Interaction with tadafil lasts up to 48 hours
Hypersensitivity
Hypotension or uncorrected hypovolaemia
