Activated Ubiquitin is attached to the second enzyme by at a cysteine residue of the enzyme
E2 transfer the Ubiquitin to the E3
Ubiquitin Protein Ligase (E3)
Attached the Ubiquitin to the lysyl amino acid residue on target protein
Protein is tagged by ubiquitin
