inhibit DNA-dependent RNA polymerase

SEs = hepatotoxicty, fever, orange urine

NB in TB

never as mono therapy: resistance

anti-TB antibiotics

isoniazid - inhibits cell wall synthesis

rifampin - inhibits transcription

pyrazinamide - inhibits synthesis of short chain FA precursors
