conditional deletion of the scf from the haematopoietic cells, osteoblasts, Nestin-Cre, or Nestin-CreER-expressing cells there were no change in the hsc function and frequency
but when scf is deleted from endothelial cells or lep r expressing perivascular cells the hscs were depleted
the niche
-
osteoblasts express many factors that promote HSC maintenance , including SCF, CXCL12, Angiopoietin-1, and Thrombopoietin
in bone marrow the hscs localize near the perisinusoidal blood vessels, thus hsc niche is perivascular
perivascular stromal cells that secrete high level of cxcl12 and nestin expressing cells that localize adjescent to blood vessels in BM the regulates hsc maintainance.

expressed by endothelial cells, bone marrow fibroblasts, osteoblasts, Cxcl12-expressing perivascular stromal cells, and Nestin-expressing mesenchymal stem cells

HSCs do not require SCF from Nestin+ cells
Scf expression by Nestin-Cre-expressing or Nestin-CreER expressing perivascular cells is not required for the maintenance of HSCs in adult bone marrow.

hsc reside s in a perivascular niche which the endothelial and the lep r expressin perivascular endothelial cells express scf for the maintenance of the BM hscs. during the developmental stage also the ter 2 marked endothelial cells with scf expression are required for the hsc maintenance and expanssion.
Perivascular stromal cells are likely
heterogeneous and may include multiple cell types like the lep r+ cells with cxcl2 or the scf that contribute to HSC maintenance
through different mechanisms.
