Pharmokinetics
Dosing
-
Drug swing: when plateau is reached, measure of difference in peak and trough.
-
Loading dose: administer much greater initial dose in order to reach maintenance/steady state sooner. Must be followed by continuous doses/graded infusions.
Bioavalibility
-
Absolute bioavailability: compares oral administration with IV administration. Assumes IV provides 100% of the available drug dose.
Relative Bioavailability: Used for oral administration and is linked to the equation for absolute bioavailability.
Distribution
Two compartment model
Main compartment for initial drug localization and second compartment for later distribution of drug
Biphasic curve when plotted on log scale, Follows first order kinetics.
