the binding of a substrate changes its quaternary structure ─ same for allosteric inhibitors and activators.
Allosteric Inhibitor
: reduces ES binding affinity.
: decreases the reaction rate (velocity)
: responsible for feedback inhibition
: stabilises the T-form (low activity)
Allosteric activator
: increases ES binding affinity.
: increases the reaction rate (velocity).
: stabilises the R-form (high activity)

At any given time, some enzymes need to turn on and others need to drop out.
activity must be regulated according to surrounding processes ─ more activity at the expense of other enzymes.
