Sleep deprivation is portrayed to be a form of torture. That Macbeth is unable to heal, relax, deprived of natures second course and has lost all innocence. Sleep signifies innocence, "the innocent sleep."

Context
During this scene, Lady Macbeth becomes involved with Macbeths ambition (when she frames the grooms) which eventually drives her mad.

Plot
Lady Macbeth drugged the grooms and Macbeth kills the kings. Macbeth fears he has murdered sleep and won't be able to sleep again. Lady Macbeth returns the daggers to Duncans chamber and tells Macbeth to stop being cowardly. The go bed after startled by knocking.
