Polypeptides of most proteins are folded up into a globular shape
Sequencing of amino acids determine the type of folding and thus the conformation of proteins
Conformation will be the same when polypeptide with particular amino acids sequencing is formed
Intramolecular bonds between amino acids in polypeptide (during folding) keep structure stable
Heat causes vibration, breaking the intermolecular bonds and changing the conformation
This is irreversible (eg. Egg white solidifying upon heating due to albumin proteins insolubility in new conformation)
If pH is deviated too far from optimum, conformation changes as intermolecular bonds break
