Isolation
Mental Issues
In Mars, you could have several mental issues like for example hallucination, Alzheimer, dementia. There is scientific proof that if you have a person locked in a box with no food, water, they start to have mental problems.
Alone
You would be completely alone when you are on Mars. No family, friends, privacy. You would have to deal with your emotions because you would be isolated from the world.
Safety
The safety in Mars is tricky because if something goes wrong it will be the agencies responsabilities.
Depressed
Do to this loneliness you could suffer from a disease called depression. You could get suicidal because of that, and NASA would not want to explain that to people.
Fear + Anxiety
Lack of oxygen
The lack of oxygen is really risky because there is a possibility that your mask would fall and then you would die of asphyxiation.
There could also have the possibility that your mask would run out of oxygen because you didn't recharge it, so you would also be suffocated

Watching Earth fade away
When you enter in space you will also see Earth fade away, and you will see all your family there and then you would be entering into the vacuum of space
When you watch Earth fade away you would see that your whole family is safe, protected by the atmosphere, magnetic field and you will not be as safe as them.
Entering into Space
When you enter in space you are entering in a vacuum that no one has ever seen. You are watching an infinite path. That must be scary.
You are going to somewhere no one as ever thought about of going, you will obviously get scared. You could have panic attacks and have anxiety.
Quality of life
Fresh Air
When you are on another planet, there will not be the level of oxygen that Earth has. So due to that astronauts when they leave biosphere 3 they would have to use a mask. Astronauts would not breath fresh air for over two years.
Mars's atmosphere is composed mostly of CO2, and Carbon Dioxide is not healthy as oxygen, on Earth the plants breath our CO2 and they give us Oxygen but how in Mars there are no plants it would be hard for people to live there.
Sleeping
When humans go to a place they never went before, it is normal for humans to have a reaction called fight or flight mode, this reaction will make you have a hard time sleeping because you will be away from your family, with a group of strangers that you don't even know, and you would need to prepare to be attentive.
You could have homesickness, that would cause trouble sleeping because you would imagine your whole family on a totally different planet than you. You could have trouble sleeping because you would not be safe or comfortable.
Privacy
In mars, there is barely enough privacy. First of all the people would not choose who they would live with, they would not choose who they would live with for 500 days.
For privacy, they had a small room that barely fits the room. The astronauts would have to stay in a tight space with almost no privacy because the biosphere three cannot be too long.
