Boil the leaf

This makes the leaf softer and breaks down the cell membrane

Boil in ethanol

This breaks down the leaf and removes the chlorophyll so there is no pigment left in the case of a colour change

Wash leaf in cold water

This softens the leaf some more as the ethanol made it very brittle it also helps to get water into the plant cells so the iodine will diffuse much more easily

Add iodine solution
