Drugs that inhibit synthesis of folic acid
Sulfonamides (sulfa drugs)
Sulfamethoazole
Competes w/ PABA to take its spot in folic acid synthesis and messes it up
Usually used for UTIs
Adverse effects
Hypersensitivity (rash, photosensitivity, SJS)
Hemolytic Anemia
Kernicterus: neurological deficits due to bilirubin in the brain
Pretty much a CYP450 INHIBITOR
Trimethoprim
TX of acute uncomplicated UTIs
Messes with a section of the folic acid synthesis
Adverse Effects
Fetal malformation
GI disturbances
Hematologic issues
Itching/rash
HyperKalemia
Trimethoprim/Sulfamethoxazole (Bactrim)
COMBO-combines mechanisms of both drugs
TX of UTI and Pneumocystis pneumonia
Interferes and multiple steps of folic acid synthesis
Adverse effects
Fetal Malformation
GI disturbances
Hemolytic anemia
Hypersensitivity
Itching/rash
HyperKalemia
